What are you trying to accomplish?  By default in Debian freshclam is run as a 
daemon and regularly updates the virus signatures.  If you want to run it by 
hand for some reason, you'll need to stop the daemon first:

sudo service clamav-freshclam stop

Don't forget to start it again afterwards or your system will get out of date.
